Dihydroceramide was Highly Elevated by the Fumonisin B1 and Desipramine in Sphingomonas chungbukensis

چکیده
− The sphingolipid metabolites act as lipid mediator for cell proliferation and apoptosis in mammalian cells. In bacteria, sphingolipid metabolism remains unknown. The purpose of this study was to investigate whether sphingolipid metabolism is potential target for fumonisin B1(FB1) and desipramine in Sphingomonas chungbukensis, Gram-negative bacteria, by comparing the intracellular contents of bacterial sphingolipids with ones of HIT-T15 β-cells, hamster pancreatic cells. The concentrations of ceramide and dihydroceramide were 18.0±12.0 and 0.025±0.018 nmol/mg protein, respectively, in HIT-T15 cells. However, the concentrations of ceramide and dihydroceramide in the bacterial culture were 2.0±1.2 and 10.6±5.5 nmol/mg protein, respectively. FB1 decreased the level of ceramide from 18.0 to 3.8 nmol/mg protein in HIT-T15 β-cells. However, dihydroceramide content in FB1-treated HIT-T15 cells was slightly decreased compared with the control culture. When S. chungbukensis was treated with either FB1 or desipramine, dihydroceramide level was increased by 5and 4-fold, respectively, compared with the control bacteria. These results indicate that FB1 and desipramine may act as an activator in bacterial sphingolipid biosynthetic pathway, and bacterial sphingolipid metabolism pathway appears to be different from the pathway of mammalian cells.

Introduction: Fumonisin B1 is a mycotoxin which can lead to neural defects, esophageal cancer, liver and kidney diseases. The aim of this study was to investigate the ability of nanocellulose conjugated with poly-lysine to remove fumonisin B1 in the different foodstuffs. Methods: First, nanocellulose was synthesized by acid hydrolysis, modified by citric acid, and conjugated with poly-lysine b...
متن کاملSurvey of fumonisin B1 contamination of cornin Northern Iran during 2000
Fumonisin B1 (FB1) is the most abundant of the fumonisin mycotoxins, mainly produced in corn by fungi of the genus Fusarium. FB1 has been shown to be hepatocarcinogenic and nephrocarcinogenic in animals. Contamination of corn with FB1 was assayed in samples collected from Mazandaran Province, situated on the Caspian littoral of Iran, in September 2000. In this survey, 38 corn samples were analy...

متن کاملPolyphasic assignment of an aromatic-degrading Pseudomonas sp., strain DJ77, in the genus Sphingomonas as Sphingomonas chungbukensis sp. nov.
Pseudomonas sp. strain DJ77, which was described as an aromatic-degrading bacterium, was investigated to clarify its taxonomic position. Phylogenetic analysis based on 16S rDNA sequences indicated that the strain belonged to the genus Sphingomonas and formed a monophyletic clade with the type strains of Sphingomonas chlorophenolica, Sphingomonas herbicidovorans and Sphingomonas yanoikuyae with ...
متن کاملPolarized membrane traffic and cell polarity development is dependent on dihydroceramide synthase-regulated sphinganine turnover.
Sphingoid bases have been implicated in various cellular processes including cell growth, apoptosis and cell differentiation. Here, we show that the regulated turnover of sphingoid bases is crucial for cell polarity development, i.e., the biogenesis of apical plasma membrane domains, in well-differentiated hepatic cells. Thus, inhibition of dihydroceramide synthase or sphinganine kinase activit...
